About the project:             

 

TET (ten eleven translocation proteins) oxidize 5mC sequentially to 5hmC, 5fC and 5caC. The 5fC and 5caC intermediates are then excised by base excision repair (BER) and other DNA damage repair (DDR) pathways. In collaboration with Prof. T Jurkowski we hope to understand sequence and modification specificity of TETs, and to apply a chemical biology approach to bring TET activity under specific small molecule control. We also plan to clarify biochemically how TETs recruit DNA repair proteins, and to resolve genetically which pathways are actually involved (SP- versus LP-BER, NER, ncMMR). Finally, we want to follow up on the controversial suggestion that DNA damage or the associated DDR proteins may recruit TETs, rather than the other way around.
